Potensi Muncul Bakteri Fotosintetik Asal Tanah Lumpur dengan Sumber Karbon dan Sulfur dalam Kolom Winogradsky

Abstract

Photosynthetic bacteria are microbes that are capable of anoxygenic photosynthesis. These bacteria can use a carbon source in the photosynthetic process. One method that can be used to determine the presence of photosynthetic bacteria is the Winogradsky column. Winogradsky column is an enrichment culture that is usually made by filling a transparent cylinder with soil or sediment and incubating it in light. The purpose of this research was to determine the photosynthetic bacteria that have the potential to appear from mud using the Winogradsky column. The research method used was descriptive qualitative and Winogradsky column. The results obtained that the carbon source with wheat flour had a red color in the water and a red layer was formed on the surface of the water. Then, the sulfur source with egg yolk has a brown color in the water with a dark brown layer on the water surface and a dark brown surface layer on the soil. The red and brown colors indicate the potential for photosynthetic bacteria to appear from the muddy soil.

 

Abstrak

Bakteri fotosintetik merupakan mikroba yang mampu melakukan fotosintesis anoksigenik. Bakteri ini mampu menggunakan sumber karbon proses fotosintesis. Salah satu metode yang dapat digunakan untuk mengetahui adanya bakteri fotosintetik adalah kolom Winogradsky. Kolom Winogradsky adalah kultur pengayaan yang biasanya dibuat dengan mengisi silinder transparan dengan tanah atau sedimen dan diinkubasi dalam cahaya. Tujuan penelitian ini adalah untuk mengetahui mengetahui bakteri fotosintetik yang berpotensi muncul terdapat di dalam tanah lumpur dengan menggunakan kolom Winogradsky. Metode penelitian yang digunakan adalah deskriptif kualitatif dan kolom Winogradsky. Hasil penelitian menunjukkan bahwa sumber karbon dengan tepung terigu terdapat warna merah pada air dan terbentuk lapisan berwarna merah pada permukaan air. Kemudian, sumber sulfur dengan kuning telur terdapat warna cokelat pada air dengan lapisan berwarna cokelat tua pada permukaan air dan lapisan permukaan tanah berwarna cokelat gelap. Warna merah dan cokelat menunjukkan potensi muncul bakteri fotosintetik dari tanah lumpur.
